Two-color Flow Cytometric Analysis of Peripheral Blood Lymphocyte Subsets in Patients with Gastrointestinal Cancer



Abstract
To elucidate alteration of peripheral blood lymphocyte subsets in patients with gastrointestinal(GI) cancer, the author analyzed T cell subsets with anti-Leu4, andti-Leu3a, anti-Leu2a, anti-Leu8 and anti-Leu15 monoclonal antibodies using
two-color
flow
cytometry in 38 cases of GI cancer including stomach cancer, hepatocellular carcinoma, and colorectal cancer as well as 18 cases of adult healthy subjects. The percentage and absolute number of CD3+cells were significantly decreased or tended to
be
decreased in patients with GI cancer, and the percentage or absolute number of CD4+cells were significantly decreased or tended to be decreased in patients with GI cancer as compared wit control. The percentage and absolute number of inducer T
cells
(Leu3a+Leu8+) were significantly decreased or tended to be decreased in patients with GI cancer, and the percentage and absolute number of helper T cells(Leu3a+Leu8-) showed no significant differences between patients with GI cancer and control.
The
percentage and absolute number of cytotoxic. T cells (Leu2a+Leu15+) were significantly decreased or tended to be decreased in patients with GI cancer, and the number of suppressor T cells (Leu2a+Leu15-) was significantly decreased or tended to be
decreased in patients with GI cancer as compared with control. From these results, it is suggested that decrease of suppressor inducer T cells and cytotoxic T cells has a role in cellular immunodeficiency of gastrointestinal cancer patients.
